RP-HPLC法测定人血浆中替考拉宁的浓度 江泓颖;曹薇;陈永刚;吴金虎;王雄 【期刊名称】《广东药学院学报》 【年(卷),期】2011(27)5 【摘 要】目的 建立测定人血浆替考拉宁的HPLC法,供血药浓度监测用,指导临床安全合理用药.方法 色谱柱为Wondasil C18柱(4.6 mm ×250 mm,5μm)；流动相为乙腈-0.05 mol/L KH2 PO4溶液(体积比28∶72,pH=3.2),检测波长为240 nm,流速为1.0 mL/min；柱温为25℃.结果 替考拉宁在4～48 μg/mL质量浓度范围内线性关系良好,检测限为2 μg/mL,方法回收率平均值为91.43％,提取回收率平均值为88.40％,日内、日间RSD值均小于6％.结论 该方法操作简单快速,准确度高,适用于替考拉宁的血药浓度测定.%Objective To establish an HPLC method for determination of teicoplanin in serum, providing the basis for monitoring clinical plasma concentrations and rational administration. Methods HPLC was performed on a C18 column (Wondasil,4. 6 mm × 250 mm,5μm) with acetonitrile-0.05 mol/L monobasic potassium phosphate 28:72 as mobile phase. The wavelength was 240 run, and the flow rate was 1. 0 mL/min, column temperature was 25℃. Results It showed good linearity in the concentration range of 4 -48 μg/mL,and the limit of quantitation was 2 μg/mL, The average method recovery was 91. 43% and the average extraction recovery was 88.40%. The inter-day RSD and intra-day RSD were both less than 6%.
